Abstract

Abstract

objective:
To detect the correlation between the activity of SLE especially lupus nephritis with ANA profile (anti-dsDNA, anti-histone and anti- nucleosome antibodies).

Methods:
Clinical and demographic data were statistically analyzed in 70 lupus nephritis patients who were diagnosed and treated during 2012–2019 in Sohag. We used serum test Euroline ANA profile (IgG)to detected the reactivity to a list of 19 autoantibodies . Correlation of the 3 autoantibodies (anti-dsDNA, - histone and - nucleosome antibodies)with nephritis was analyzed.Renal biopsies were studied by pathologists. The Immune complex deposition was recognized by immunohistochemistry stain.

Results:
Our study detected the role of reactivity of anti-nucleosome - dsDNA and -histone antibodies in the diagnosis of lupus nephritis. Furthermore, our data suggest that we can predict the kidney affection and activity of lupus nephritis by the co-positive anti-dsDNA, - nucleosome and -histone antibodies ,which may add a major benefit in the follow-up and treatment of these patients.

Close reactivity with anti-dsDNA, -nucleosome and histone antibodies (3-positive antibodies) in SLE patients were highly relevant to Lupus nephritis pathology. Three-pos LN patients showed significantly higher serum levels of these antibodies, suffered from more severe nephropathy and needed more intensive treatments than non-3-pos LN patients, indicating 3-pos as an indicative biomarker for severe LN.

Conclusions:
ANA profile (IgG) especially the 3 autoantibodies (Anti-nucleosome - histone and -dsDNA antibodies ) maybe used as indicators for severe kidney affection and activity in SLE patients.
